- TRAF3 mediates the signal transduction from TNF receptors and Toll-like receptors, leading to the activation of NF-κB, MAP kinases, or IRF3. TRAF3 is involved in the regulation of anti-viral response and B-cell survival.

- TRAF3 forms a homodimer. TRAF3 also forms heterodimers with TRAF2 and TRAF5. TRAF3 also interacts with CD40, TICAM1, TRAFD1, BIRC2, BIRC3, TLR4. TRIP, and TBK1.
